Cycloalkyne in Reactive groups

Strained cycloalkynes, like cyclooctynes, react quickly with azides in a reaction called spAAC, strain promoted alkyne-azide cycloaddition. The reaction does not require catalyst, it is very rapid and high yielding. The process is useful for the conjugation of different classes of molecules even in vivo.

There is a balance between reactivity and stability of cycloalkynes. Azodibenzocyclooctynes (DBCO, ADIBO) are cyclooctyne derivatives that are characterized by excellent reaction kinetics, and very good stability.
