Nuclear stains
Nuclear stains are fluorescent probes that selectively label cell nuclei by binding to nucleic acids, primarily DNA, via interaction with the minor groove of DNA (often with a preference for A—T-rich regions) or intercalating between adjacent base pairs. Upon binding, the fluorescence intensity of these dyes increases dramatically, providing high signal-to-background contrast and clear nuclear localization.
These dyes are essential tools in fluorescence microscopy, flow cytometry, and high-content imaging, enabling visualization of nuclear morphology, chromatin organization, and cell cycle status. Dyes that penetrate intact membranes are used as markers for live cells, while non-penetrating dyes serve as markers for apoptotic or dead cells.
