Tetraacetylated N-(4-pentynoyl)glucosamine (Ac4GlcNAl) is a synthetic monosaccharide labeled with an alkyne. It penetrates cells and is metabolically incorporated into glycoproteins in place of natural N-acetylglucosamine (GlcNAc).
For the subsequent detection of modified glycoproteins, a Cu(I)-catalyzed (CuAAC) click reaction using azides or biotin-azide is employed.
