Catalytic buffer for Click chemistry contains copper(II), triethylammonium acetate, pH 7, and DMSO. The buffer is a 1.5x concentrate, a ready-to-use reagent for the modification of nucleic acids such as oligonucleotides, or small molecules.
Copper(II) should be reduced to copper(I) prior to reaction. Ascorbic acid is the recommended reductant.
THPTA is a water soluble ligand for Cu(I) catalyzed Click chemistry. The ligand stabilizes copper in its Cu(I) oxidation state. Due to high aqueous solubility, reactions with this ligand do not require an organic co-solvent.
