Singlet Oxygen Sensor Green (SOSG) is a fluorogenic probe for detecting singlet oxygen (1O2) in aqueous solutions. SOSG is able to incorporate into a living mammalian cells and can be used for in vivo imaging of intracellular 1O2 and various clinical applications.

The SOSG molecule consists of two parts: a fluorophore and an anthracene-derived trapping moiety. In the absence of singlet oxygen, emission from the fluorophore is quenched by electron transfer from the adjacent trapping moiety. The indicator exhibits weak blue fluorescence with excitation/emission maxima at 372/395 and 393/416 nm. In an environment containing 1O2, the trapping moiety reacts with singlet oxygen and forms an endoperoxide anthracene, which is no longer an efficient intramolecular electron donor. Removing the quenching leads to the fluorophore’s fluorescence at excitation/emission maxima of about 504/525 nm.

SOSG reaction

In contrast to other reagents for 1O2 detection, SOSG is highly selective for singlet oxygen and does not show a noticeable response to superoxide (•O2−) or hydroxyl radical (•OH). However, it is activated at alkaline pH and in certain solvents, such as acetonitrile, DMSO, DMF, and acetone.

Upon receipt, store the reagent desiccated and protected from light at −20°C, do not open the tubes until use. To prepare a stock 5 mM solution, dissolve the contents of 100 µg tube in 33 µl of methanol. Prepare working solutions of SOSG immediately before use and discard any excess diluted reagent at the end of the work session. The optimal working concentration of SOSG should be determined empirically; a suggested starting concentration range is 1–10 µM.
